Is there a difference between firefly luciferin, beetle luciferin, and D-Luciferin from Cellcyto?
No. D-Luciferin from Cellcyto, firefly luciferin, and beetle luciferin are all names for the same chemical, (S)-2-(6-Hydroxy-2-benzothiazolyl)-2-thiazoline-4-carboxylic acid. We only provide D-Luciferin Potassium Salt for in vivo and in vitro analysis.
For what applications is Cellcyto D-Luciferin suitable?
Cellcyto D-Luciferin is used in all luciferase applications including gene reporter assays, hygene testing, sanitation monitoring, pyrosequencing, and whole animal imaging.
How is D-Luciferin prepared for whole animal imaging?
D-Luciferin salts are easily soluble up to 30mg/mL in unbuffered water and many preps simply dissolve the luciferin potassium salt at 3-15 mg/mL for i.p. administration.
If a buffering system is to be used, pH and time are very important as D-Luciferin is relatively unstable at low pH (<6.5, hydrolysis) and high pH (>7.5, conversion to L-Luciferin).
What is the difference between the free acid, sodium salt and potassium salt?
The main difference between the free acid and the salt forms is the solubility and dissolution characteristics. D-Luciferin free acid has very low solubility in water and dissolves very slowly into buffered solutions. The potassium salts dissolves quite easily and quickly into water.

How stable is D-luciferin in solution?
Frozen (-80°C) samples have been reported to be suitable for use after a year. Liquid samples at room temperature show degradation of up to 0.25% per day with the degradation route being pH dependant. If oxygen is present in the solution, dehydroluciferin will form.

2、D-荧光素应用于活体成像的基本原理
细胞中D-荧光素酶参与的化学发光反应遵守酶促反应的基本原理,在底物D-荧光素钾过量的情况下,反应产生的光子数(发光量)与细胞中的D-荧光素酶的量成正相关,从而可以推算出细胞的数量。
3、D-荧光素钾盐溶液的稳定性
D-荧光素钾盐易溶于水和缓冲液,溶解度可高达25mg/mL。一般使用浓度为3-15 mg/mL。溶液的pH值、溶液中的氧气和保存时间对其保存过程中的稳定性非常重要。当溶液的pH<6.5(发生水解作用)或>7.5(发生消旋化作用,D型转化为L型)的情况下,D-荧光素钾盐相对不稳定。如果溶液中存在少量的氧气,将加速D-荧光素钾的降解速度。
D-荧光素配成溶液后,在最佳的保存条件(-80°C,最佳pH,无氧气)下,存在固定的降解速度:0.2%/天,因此配成溶液保存的D-荧光素应尽量在1年内使用完。
有水存在的情况下,主要发生消旋化作用,即D-荧光素转化成L-荧光素,有报道称L-荧光素对荧光素酶催化的酶促反应有抑制作用。

主要区别在于溶解和分散性能。D-荧光素钾盐在水和缓冲液中的溶解度高,且溶解速度较快,故在一般的生物学试验中常用D-荧光素钾盐。
5、D-荧光素钾盐的保存和运输条件
保存条件:≤-20°C,避光,保持干燥。(在不打开包装的条件下,可保存两年时间)。
运输条件:高纯度的D-荧光素钾盐可在短时间内常温条件下保存,运输条件为常温。
原装(没有打开包装)荧光素的包装瓶中充有氩气,保证其稳定性,打开包装后,请严格按照保存条件保存。
D-荧光素钾盐具有易溶于水的特性,其暴露于空气中,易吸潮。在称量过程中,请考虑其易吸潮的特性,先进行温度平衡,再快速称取。
